N2 - Chronic hepatitis C disease causes fibrosis, cirrhosis, and liver cancer, resulting in abnormal liver function cause mortality. The development of hepatitis C therapy in the last decade has resulted in severe side effects, resistance, and high cost, thus is needed as an alternative to medicinal plants. Gedong Gincu cultivar is a typical mango plant (Mangifera indica L.) from the Cirebon region which has characteristics such as aroma, taste, and skin color as well as its nutritional content and with high economic value compared to other mango cultivars. The aim of this study was to screen the Gedong Gincu cultivar mango plant leaves, fruit pulp, seeds and fruit skin against Hepatitis C Virus (HCV). In vitro study used the Huh7 cell line to examine the gedong gincu plant ethanol extract against HCV is used a focus on forming units for anti-HCV and MTT assay to examine the cytotoxicity against the Huh7 cell line. The results showed that four extracts, ethanol extracts from leaves (EL), fruit pulp (EFP), seeds (ES), and fruit skin (EFS) of the Gedong Gincu cultivar mango, with inhibition concentration 50% (IC50) of 58.8, 110.4, 8.2, and 27.4 μg/mL, respectively. Huh7it cells were incubated with the extracts for 48 h and it appears that EL, EFP, ES, and EFS is not toxic up to 800 μg/mL. These results suggested that ES of Mangifera indica gedong gincu cultivar is a potential source of anti-HCV and required to isolate the antiviral compound as anti-HCV.
