Abstract

The aim of this study was to measure the effect of gabapentin administration on the Calcitonin Gene-Related Peptide (CGRP) and interleukin-10 (IL-10) in experimental animals with models of peripheral nerve neuropathic lesions. This work was an experimental research using 23 Rattus norvegicus strain Sprague-Dawley with peripheral nerve neuropathic lesion model achieved using a partial sciatic nerve ligation (PSL) procedure. The experimental mice were separated into 3 groups, namely the normal group, the PSL group without Gabapentin and the PSL group with Gabapentin. Laminectomy was done to obtain the spinal cord at the level of the ischiadic nerve roots specimen. The specimen was stained with immunohistochemistry (IHC), and then the expression of the CGRP and IL-10 was calculated using the immunoreactive score (IRS). The results showed that there was a significant difference in CGRP expression between groups (p=0.001). When the results were analyzed using Mann Whitney analysis, there indeed was substantial difference between the treatment group and the positive control group (p=0.002). In calculating the expression of the anti-inflammatory cytokine IL-10, there was no remarkable difference between groups, meaning that the result was not statistically significant (p=0.85). The administration of Gabapentin can reduce CGRP expression. However, it can not increase the expression of IL-10 in the dorsal horn of the medulla spinalis after neuropathic lesions of the peripheral nerves.
