Effect of freeze dried bovine bone xenograft particles on runx-2 expression in human bone marrow mesenchymal stem cell

Abstract

– Xenograft materials can be obtained from cow bone or called bovine-derived bone xenograft (BDX). Freeze Dried Bovine Bone Xenograft (FDBBX) is produced by freeze drying so that the remaining part consist of inorganic and organic material in which biological molecules are still retained. Hence, FDBBX theoretically has osteogenic induction potential. The aim of this study is to analyze RUNX-2 expression in human bone marrow mesenchymal stem cells (hBM-MSC) culture after induction with FDBBX-conditioned medium. This study was carried out using hBM-MSC culture which was divided into experimental and control groups. In experimental groups, hBM-MSC was culture in 2.5% FDBX-conditioned medium while basic medium was used in control group. After 1, 3, and 5 days of culture, the cells were fixed and immunostained to evaluate RUNX-2 expression. The result of study is no RUNX-2 expression noted in control group. There is constant increase in the number of cells expressing RUNX-2 on hBM-MSC culture in experimental groups. So, Freeze Dried Bovine Bone Xenograft has osteogenic induction potential in vitro.
